要旨

The genomes of complex eukaryotes contain vast intergenic regions that are mostly composed of repetitive DNA, with transposons as a major constituent. Although it is now apparent that some transposons and pseudogenes participate in gene regulation, the function of the majority of these sequences remains unknown. Piwi proteins regulate retrotransposons for degradation using piRNAs as guides in the germline across the animal kingdom. We recently found that piRNAs mediate the degradation of a large number of mRNAs and lncRNAs in the mouse germline. In this regulation, retrotransposon and pseudogenes function as regulatory sequences, and retrotransposon sequences in the 3´UTRs of mRNAs are targeted by piRNAs for degradation. Many genes harbor retrotransposon sequences in their 3´UTRs and promoters. To find novel retrotransposon regulators potentially involved in the regulation of such genes, I am planning to perform RNAi screening.
