要 旨

Heart failure is associated with high levels of DNA damage and defective repair due to oxidative stress. 8-oxoguanine (8-oxoG) is one of the most common DNA lesions in heart failure and its presence has been correlated to mutations, apoptosis and mitochondrial dysfunction. The principal base excision repair enzyme responsible for repair of this DNA damage is OGG1 and its expression has been shown to be reduced in heart failure. We are elucidating the role of OGG1 in stress-induced heart failure and repair by using Ogg1-/- mice. Heart failure in the mice is induced using a pressure-overload model of transverse aortic constriction (TAC) and we compare the progression of heart failure in the knock out mice with wild type mice. First we are employing RNA-Seq and qPCR to look at the effect of OGG1 loss on cardiac transcriptome. Cardiac Echocardiography is used to assess important physiological parameters such as ejection fraction, fractional shortening and wall dimensions. In addition histological stains are used to evaluate fibrosis and hypertrophy.
